Working as a microbiologist of the specimen is not stained I will start with gram staining of the specimen and if the specimen was stained then I will proceed to observation..In case there are any host cells such as white blood cells or the epithelial cells are present in the sample ,if gram stained they are positive for the gram stain which imply that the white blood cells are infected with the gram negative bacteria and if the gram stain is negative then they are infected with the gram positive bacteria.The observation is similar with the epithelial cells.

Gram positive bacteria includes Bacillus anthracis and they have a thicker peptidoglycan cell wall which has 20-80 nm thick while the gram negative bacteria has a thin cell wall 2-3nm thick.Example of gram Negative bacteria includes Escherichia coli.

.If the gram stain and the cells turns to purple then they are gram positive bacteria meaning no infection in the host cells like the epithelial cell and white blood cells or it implies the presence of gram positive bacteria.If cells turn pink colour after staining then they are gram negative hence the epithelial cells and white blood cells are infected with the gram negative bacteria that causes the infection.
